![]() ![]() In the absence of catalase, neither EQ nor superoxide dismutase (SOD), an alternative antioxidant, were able to afford protection of the myocardium. In the presence of catalase or EQ, CK release was reduced and infarct size was decreased. Protection was also afforded by the antioxidant ethoxyquin (EQ), as assessed both in vitro and in vivo. It was demonstrated that the free radical scavenger catalase could protect the myocardium from reperfusion-induced damage by preventing lactate production. ![]() In the in vivo model of reperfusion, creatine kinase (CK) release and infarct size were assessed. Lactate production and oxygen consumption were measured by flow-injection analysis. H(2)O(2) was used as a free radical inducer. Catalase was added to the medium as an antioxidant. The role of free radicals was demonstrated in an in vitro study, where myocardial oxygen consumption and lactate production were evaluated in a cell culture medium. Myocardial oxygen consumption and lactate production were assessed in the isolated perfused rat heart using microdialysis technique to evaluate the vulnerability of the tissue to ischemia. We therefore investigated whether antioxidants could prevent or attenuate ischemia/reperfusion damage. The extracellular generation of free radicals in ischemic tissue may be harmful to the cell, and a radical scavenger may be beneficial by limiting the extent of the damage.
